Lecture Summary

We will introduce how to use fluorescence technology-based two-dimensional electrophoresis to improve the detection and analysis of low-abundance tyrosine phosphorylated proteins. To investigate the consistency of fluorescent Western blotting and fluorescent differential electrophoresis (2-D DIGE) in the detection of tyrosine phosphorylated proteins that were not enriched or enriched by immunoaffinity chromatography. A model system with known changes in phosphorylation levels in cells treated with Imatinib mesylate was used to test the effectiveness of these new methods in detecting changes in tyrosine phosphorylation levels. The results show that enrichment of related proteins prior to analysis is more helpful for accurate determination of results, and we obtained the results of processing samples and control samples in a simplified 2-D DIGE experiment (after enrichment by immunoaffinity method) The results of differential expression of tyrosine phosphorylated proteins between samples) were also identified using mass spectrometry.
